Key Takeaways
- ✓Moderna valuation context: Moderna's market cap reached nearly $100 billion before the cancer vaccine data leak, surpassing Sanofi at $90 billion despite only $2.2–2.3 billion in annual revenue. When analyzing biotech stock moves, measure from the original trial success announcement rather than the prior day's price — doing so reveals the stock remained well above pre-announcement levels, making "crash" headlines misleading and context-dependent.
- ✓Options distortion in biotech markets: Retail traders holding short-dated options on acquisition targets like Abivax systematically distort price discovery and public sentiment. Because time premium erodes continuously, options holders need stocks to move immediately or face total loss — creating panic on normal pullbacks. Investors tracking sector health should monitor options-to-stock ownership ratios as a sentiment distortion signal, separate from fundamental analysis.
- ✓Degrossing as a structural market risk: Long-short platform funds, which have not had a down year in roughly a decade, now represent enough capital in biotech to cause sector-wide repricing through degrossing events unrelated to fundamentals. When heavily shorted names like Iovance and Twist spike, forced selling of long positions follows. Operators and investors should factor fund-flow technicals alongside data readouts when interpreting sudden sector-wide valuation swings.
- ✓China licensing as pharma's pipeline alternative: Roche, Gilead, and Eli Lilly each completed China-sourced licensing deals in the same week — Gilead paying $1.3 billion upfront for an oral IL-23 peptide. Pharma companies are increasingly bypassing US biotech acquisitions by licensing directly from Chinese biotechs. Biotech executives should monitor whether the US Treasury's proposed investment restrictions, currently in public comment, will accelerate this trend by penalizing venture-backed Chinese startups more than direct pharma licenses.
- ✓Silence Therapeutics turnaround blueprint: Ali Mortazavi rebuilt Silence Therapeutics from a £5 million market cap by abandoning a failing oncology program, pivoting from lipid nanoparticles to GalNAc delivery after benchmarking Alnylam, and taking a 10% stake in Arrowhead at a 70% discount to cash. The eventual $1.3 billion Merck KGAA acquisition at a 130% premium was driven primarily by divisiran targeting polycythemia vera — an indication switch made by a subsequent management team, illustrating that platform pivots compound across leadership generations.
What It Covers
Biotech Hangout's 200th episode, recorded October 23, 2026, brings together seven co-hosts and Silence Therapeutics former CEO Ali Mortazavi to examine Moderna's cancer vaccine stock volatility, the Merck KGAA acquisition of Silence Therapeutics for $1.3 billion, bearish biotech market dynamics, pharma CEO social media engagement, and five-year industry retrospectives from veteran operators and investors.
Key Questions Answered
- •Moderna valuation context: Moderna's market cap reached nearly $100 billion before the cancer vaccine data leak, surpassing Sanofi at $90 billion despite only $2.2–2.3 billion in annual revenue. When analyzing biotech stock moves, measure from the original trial success announcement rather than the prior day's price — doing so reveals the stock remained well above pre-announcement levels, making "crash" headlines misleading and context-dependent.
- •Options distortion in biotech markets: Retail traders holding short-dated options on acquisition targets like Abivax systematically distort price discovery and public sentiment. Because time premium erodes continuously, options holders need stocks to move immediately or face total loss — creating panic on normal pullbacks. Investors tracking sector health should monitor options-to-stock ownership ratios as a sentiment distortion signal, separate from fundamental analysis.

The eventual $1.3 billion Merck KGAA acquisition at a 130% premium was driven primarily by divisiran targeting polycythemia vera — an indication switch made by a subsequent management team, illustrating that platform pivots compound across leadership generations.
- •Pharma CEO direct social media engagement: Eli Lilly CEO Dave Ricks posting unfiltered content on X without legal or communications review is cited as a model for rebuilding public trust in pharma. Brad Lunkar confirmed firsthand that Ricks writes his own posts. Given that pharma ranks among the most distrusted industries publicly, executives who demonstrate technical knowledge and personal authenticity on social platforms can shift perception more effectively than traditional corporate communications — and the CEO of the largest pharma company doing so removes compliance objections for others.
Notable Moment
Ali Mortazavi revealed that Silence Therapeutics sold a 10% stake in Arrowhead — purchased when Arrowhead traded at a 70% discount to cash — before it appreciated to a value of $900 million within four to five months. The board sold for a modest profit to extend runway, illustrating how survival decisions routinely cost biotech companies transformative upside.

You're listening to Biotech Hangout, a live and unedited weekly discussion of all the latest news in our industry with a group of biotech leaders and experts. I'm Daphne Zohar, and my co hosts today are Mack Line, Eric Schmidt, Chris Garabedian, Brad Lunkar, Sam Fazeli and Josh Schimmer. We're also joined by special guest Ali Mortazavi, former CEO of Silence Therapeutics, which was in the news this week. For more information about our hosts and guest speakers or to listen to the most recent episode, please go to biotechhangout.com. Think Ali, can you go on mute please? Sure. Thanks. Today is the two hundredth recorded episode of Biotech Hangout with the first recorded episode five years ago in December 2021 and that was with Chris, Brad and I hosting. Looking back on that day John Mergonori joined the show after announcing his transition from Alnylam. And we had Damian Garde and Adam Forstein of STAT talking through the reporting on Biogen's Aguhelm, which was one of the big stories of the year. However, that actually wasn't our first episode. Biotech Hangout, which was formerly known as Clubhouse, began as a closed door session in January 2021 after the first virtual JPMorgan conference. And I don't think I was at that one, but I joined soon after. Since then, we've had over 60 newsmaking guests, and our top episodes have had more than 15,000 listeners tune in across platforms. So Hangout is basically like the water cooler of what the industry is talking and thinking about, and what our listeners may not know is that it's entirely unscripted. We schedule the hosts and then agree on topics a few days before the show. Our hosts prep their own topics, but it wouldn't be possible without Shannon Costello and Daniela Spector who do everything before, during, and after to make sure the show runs smoothly and episodes are available to all. So we're going to go around quickly with some thoughts on Hangout, what makes it unique, and what are you hearing from listeners. And I'll start with my Hangout cofounders, Chris and Brad. Chris, start with you. Yeah. Great. Well, Daphne, congrats to you as well for taking that initial idea on Clubhouse, which was a little bit more of a party line. Like, people could come on. They could ask questions. It was a little bit of a free for all. That was kind of the clubhouse format. And, you know, when you moved it over to Twitter, now x spaces, you made the really good decision to broaden the cohosts, right, you know, have a rotating group of folks who could talk about any topics, still have guests on occasion like Ali today. And so it's it's amazing that, you know, we've been, you know, five years plus running strong, and, and it does take a lot of work. I think when Brad and I kinda took a little bit of a a backseat to be involved, you …
